Can I raise my GPA significantly in one semester?

It depends on how many credits you have already completed. Early in your degree, a strong semester can move the GPA by 0.2–0.3 points. After 90 credits, 15 new credits can realistically only shift the cumulative GPA by about 0.05–0.1 points per semester even with straight A's.

What if my school uses a scale above 4.0?

Some schools award 4.3 for A+ grades, giving a theoretical maximum above 4.0. If your school does this, your needed term GPA result above 4.0 may actually still be achievable. Confirm your institution's exact scale with the registrar.

What does it mean if the needed term GPA is negative?

It means your current GPA is already above the target, and no matter how poorly you do this term (while still passing), your cumulative GPA will remain above your stated goal. You are in a safe zone for that target.
